光电信息大讲堂第六十三讲:超声治疗 – 从冲击波碎石到单细胞分析Therapeutic Ultrasound – from shock wave lithotripsy to single cell analysis

报告摘要:
       超声治疗是工程学,生物学,临床医学交叉的新兴领域。高频率,高强度的声波能够被用于局部组织加热,靶向药物和基因传递,碎裂结石,以及在各种不同的生物医学和临床应用中对单细胞的力学调控和变形。在这次报告中,我将介绍在超声治疗中不同尺度的两个例子:1)冲击波碎石(SWL)–无损伤治疗患者体内肾结石,和 2)单细胞分析。首先,我将按时间顺序回顾在过去的三十年的研究成果,即对冲击波和肾结石,肾组织之间的相互作用,包括超声空化产生的副作用的理解。其次,我要描述我们最近的工作,通过在微流控芯片中激光产生的串联气泡和表面图案化来研究空化射流引起的涡流和细胞膜变形及由此产生的生物效应之间的因果关系。这些研究展示了工程设计和手段在维护及提高超声治疗效率以及其在生物医学和临床应用研究中所发挥的重要作用。
     Therapeutic ultrasound is an emerging field at the interface of engineering, biology and clinical medicine.  High frequency, and often high intensity, acoustic waves in MHz range can be used for localized tissue heating, targeted drug and gene delivery, fragmentation of concretions, mechanical manipulation and deformation of single cells with a diverse range of biomedical and clinical applications. In this talk, I’ll present two examples of distinctly different scales in shock wave lithotripsy (SWL) – a non-invasive technology for the disintegration of kidney stones in patients, and single cell analysis.  First, I’ll review chronologically the research efforts in the past three decades to understand the dynamic interaction of shock waves with kidney stones and renal tissue, including secondary effects produced by cavitation. Second, I’ll describe recent work in my lab using laser-generated tandem bubbles in a microfluidic chip and surface patterning to investigate the causal relationship between cavitation jetting-induced vortical flow, cell membrane deformation and resultant bioeffects. Altogether, these studies illustrate the important role that engineering plays in safeguarding and improving the efficiency of therapeutic ultrasound in biomedical and clinical applications.

   美国杜克大学材料科学与机械工程学院 Anderson-Rupp 教授,生物医学工程系教授,美国机械工程师学会(ASME)会士,美国生物医学超声/生物反应振动声学学会技术委员会委员,NIH MERIT Award 奖,美国泌尿外科疾病基金会/塞尔新研究者研究奖,杜克大学 Capers & Marion McDonald 指导建议奖,在 PNAS 和 PRL 等杂志上发表了多篇高水平论文。研究主要集中在冲击波碎石术(SWL)、超声介导药物和基因传递,高强度超声聚焦(HIFU)肿瘤治疗和超声空化的生物效应。在超声波碎石和组织损伤的基础研究中做出了重大贡献,以及提出无损伤的治疗方法来提高对尿石症患者的安全性和疗效。
     Dr. Pei Zhong is Full Professor in the Department of Mechanical Engineering and Materials Science, Biomedical Engineering at Duke University, Durham, North Carolina, with research interest primarily on shock wave lithotripsy (SWL), ultrasound-mediated drug and gene delivery, high-intensity focused ultrasound (HIFU) for cancer therapy, and bioeffects of cavitation. He is a nationally and internationally established leader in the field of SWL and therapeutic ultrasound.  He have contributed significantly to the fundamental knowledge of stone comminution and tissue injury in SWL, as well as development of innovative technologies for improving the safety and efficacy of this non-invasive treatment modality for patients with urolithiasis. As a result, he was awarded a NIH MERIT Award in 2010 for his long-lasting research efforts and contributions to SWL. He was also elected a Fellow of the American Society of Mechanical Engineering (ASME) in 2011.
